Chromosome-level genome assembly and population genetic analysis of a near-threatened rosewood species (Dalbergia cultrata Pierre Graham ex Benth) provide insights into its evolutionary and cold stress responses.

Dalbergia cultrata Pierre Graham ex Benth (D. cultrata) is a precious rosewood tree species that grows in the tropical and subtropical regions of Asia. In this study, we used PacBio long-reading sequencing technology and Hi-C assistance to sequence and assemble the reference genome of D. cultrata. We generated 171.47 Gb PacBio long reads and 72.43 Gb Hi-C data and yielded an assembly of 10 pseudochromosomes with a total size of 690.99 Mb and Scaffold N50 of 65.76 Mb. The analysis of specific genes revealed that the triterpenoids represented by lupeol may play an important role in D. cultrata's potential medicinal value. Using the new reference genome, we analyzed the resequencing of 19 Dalbergia accessions and found that D. cultrata and D. cochinchinensis have the latest genetic relationship. Transcriptome sequencing of D. cultrata leaves grown under cold stress revealed that MYB transcription factor and E3 ubiquitin ligase may be playing an important role in the cold response of D. cultrata. Genome resources and identified genetic variation, especially those genes related to the biosynthesis of phytochemicals and cold stress response, will be helpful for the introduction, domestication, utilization, and further breeding of Dalbergia species.

Proposing Signaling Molecules as Key Optimization Targets for Intensifying the Phytochemical Biosynthesis Induced by Emerging Nonthermal Stress Pretreatments of Plant-Based Foods: A Focus on γ-Aminobutyric Acid.

Emerging evidence has confirmed the role of emerging nonthermal stressors (e.g., electromagnetic fields, ultrasonication, plasma) in accumulating bioactive metabolites in plant-based food. However, the signal decoding mechanisms behind NonTt-driven phytochemical production remain unclear, hindering postharvest bioactive component intensification. This study aims to summarize the association between signaling molecules and bioactive secondary metabolite production under nonthermal conditions, demonstrating the feasibility of enhancing phytochemical accumulation through signaling molecule crosstalk manipulation. Nonthermal elicitors were found to be capable of inducing stress metabolisms and activating various signaling molecules, similar to conventional abiotic stress. A simplified pathway model for nonthermally induced γ-aminobutyric acid accumulation was proposed with reactive oxygen species and calcium signaling being versatile pathways responsive to nonthermal elicitors. Manipulating signal molecules/pathways under nonthermal conditions can intensify phytochemical biosynthesis. Further research is needed to integrate signaling molecule responses and metabolic network shifts in nonthermally stressed plant-based matrices, balancing quality modifications and intensification of food functionality potential.

Fixed vs. variable light quality in vertical farming: Impacts on vegetative growth and nutritional quality of lettuce.

Lettuce (Lactuca sativa) is commonly produced in vertical farms. The levels of nutritionally important phytochemicals such as beta-carotene (precursor to vitamin A) are generally low in lettuce. In this study, we investigated the benefits of variable lighting strategy (i.e., varying the light quality during production) on maintaining plant growth and increasing the biosynthesis of beta-carotene and anthocyanin. We tested two variable lighting methods, using green and red romaine lettuce, namely (i) providing growth lighting (supports vegetative growth) initially (21 days) followed by a high percentage of blue light (supports biosynthesis of phytochemicals) at final stages (10 days) and (ii) providing a high percentage of blue light initially followed by growth lighting at final stages. Our results indicate that the variable lighting method with initial growth lighting and high percentage of blue at final stages can maintain vegetative growth and enhance phytochemicals such as beta-carotene in green romaine lettuce while both variable lighting methods were not effective in red romaine lettuce. In green romaine lettuce, we did not observe a significant reduction in shoot dry weight but there was an increase in beta-carotene (35.7%) in the variable compared to the fixed lighting method with growth lighting for the entire duration. The physiological bases for differences in vegetative growth and synthesis of beta-carotene and anthocyanin in the variable and fixed lighting methods are discussed.

Effect of postharvest visible spectrum LED lighting on quality and bioactive compounds of tomatoes during shelf life

The objective was to evaluate the effect of visible spectrum LED lighting during shelf-life on physicochemical quality and the main bioactive compounds of Kumato® cherry tomatoes. Tomatoes were stored 13 days at 5 °C under white (W), blue (B), blue + red (B + R), green + red (G + R), and green + far-red (G + FR) LED lights. Darkness (D) was used as control. Tomatoes under illumination showed higher weight losses and firmness decreases (30–35%). No chilling injury was observed. B + R lighting increased the carotenoid content by ∼27%, while G + R and G + FR reported an increase ∼30% in phenolics. B and B + R showed the highest increase in the phytochemical biosynthesis (lycopene and naringenin, as main carotenoid and flavonoid found) compared to D and W. Conclusively, illumination with B + R or B during shelf-life is recommended to enhance the main bioactive compounds. G + R and G + FR, also reported to be good elicitors of the phenolics and carotenoids biosynthesis.

Polish Achievements in Bioactive Compound Production From In Vitro Plant Cultures

AbstractPlant cell and organ cultures are potential sources of valuable secondary metabolites that can be used as food additives, nutraceuticals, cosmeceuticals, and pharmaceuticals. Phytochemical biosynthesis in various in vitro plant cultures, in contrast to that in planta, is independent of environmental conditions and free from quality fluctuations.Pharmaceutical application of plant biotechnology is of interest to almost all departments of the Faculty of Pharmacy and Institute of Pharmacology in Poland with a botanical profile (Pharmaceutical Botany, Pharmacognosy, and Pharmacology).This study discusses the advances in plant biotechnology for the production of known metabolites and/or biosynthesis of novel compounds in plant cell and organ in vitro cultures in several scientific centers in Poland.

Effects of Post-Harvest Elicitor Treatments with Ultrasound, UV- and Photosynthetic Active Radiation on Polyphenols, Glucosinolates and Antioxidant Activity in a Waste Fraction of White Cabbage (Brassica oleracea var. capitata).

Biosynthesis of phytochemicals in leaves of Brassica can be initiated by abiotic factors. The aim of the study was to investigate elicitor treatments to add value to waste of cabbage. A leaf waste fraction from industrial trimming of head cabbage was exposed to UV radiation (250–400 nm, 59 and 99 kJ∙m−2, respectively), photosynthetic active radiation (PAR, 400–700 nm, 497 kJ∙m−2), and ultrasound in water bath (35 kHz, at 15, 30 and 61 kJ∙l−1 water), in order to improve nutraceutical concentration. UV was more effective than PAR to increase the level of flavonols (2 to 3-fold higher) and hydroxycinnamate monosaccharides (1 to 10-fold higher). PAR was three times as effective as UV to increase anthocyanins. Interaction of PAR + UV increased antioxidant activity (30%), the content of five phenolics (1.4 to 10-fold higher), and hydroxycinnamic monosaccharides (compared with PAR or UV alone). Indoles were reduced (40–52%) by UV, but the other glucosinolates (GLS) were unaffected. Ultrasound did not influence any parameters. The results are important for white cabbage by-products by demonstrating that UV + PAR can be successfully used as an effectual tool to increase important phenolics and antioxidant activity of waste fraction leaves without an adverse effect on the main GLS.

Nutrition vs association: plant defenses are altered by arbuscular mycorrhizal fungi association not by nutritional provisioning alone

BackgroundWhile it is known that arbuscular mycorrhizal fungi (AMF) can improve nutrient acquisition and herbivore resistance in crops, the mechanisms by which AMF influence plant defense remain unknown. Plants respond to herbivory with a cascade of gene expression and phytochemical biosynthesis. Given that the production of defensive phytochemicals requires nutrients, a commonly invoked hypothesis is that the improvement to plant defense when grown with AMF is simply due to an increased availability of nutrients. An alternative hypothesis is that the AMF effect on herbivory is due to changes in plant defense gene expression that are not simply due to nutrient availability. In this study, we tested whether changes in plant defenses are regulated by nutritional provisioning alone or the response of plant to AMF associations. Maize plants grown with or without AMF and with one of three fertilizer treatments (standard, 2 × nitrogen, or 2 × phosphorous) were infested with fall armyworm (Spodoptera frugiperda; FAW) for 72 h. We measured general plant characteristics (e.g. height, number of leaves), relative gene expression (rtPCR) of three defensive genes (lox3, mpi, and pr5), total plant N and P nutrient content, and change in FAW mass per plant.ResultsWe found that AMF drove the defense response of maize by increasing the expression of mpi and pr5. Furthermore, while AMF increased the total phosphorous content of maize it had no impact on maize nitrogen. Fertilization alone did not alter upregulation of any of the 3 induced defense genes tested, suggesting the mechanism through which AMF upregulate defenses is not solely via increased N or P plant nutrition.ConclusionThis work supports that maize defense may be optimized by AMF associations alone, reducing the need for artificial inputs when managing FAW.

UVA-Radiation Exposure of Different Durations Promoted the Growth, Phytochemicals and Glucosinolate Biosynthesis of Chinese Kale.

Ultraviolet-A (UVA) (315–400 nm) is an essential environmental signal that regulates plant development and affects phytochemicals biosynthesis, including glucosinolate biosynthesis. The effects of different UVA (380 ± 10 nm, 40 μmol/m2/s) exposure durations, including 0 h/d (UV0), 6 h/d (UV6) and 12 h/d (UV12), on the growth and phytochemicals of Chinese kale (Brassica alboglabra) under white 250 μmol/m2/s LEDs were investigated. UVA exposure of different durations influenced the growth and phytochemicals biosynthesis of Chinese kale. Prolonging UVA irradiation throughout the growth cycle positively affected the growth and the development of Chinese kale, with evident increases in the dry weights of shoots and roots, plant height, stem diameter, specific leaf weight and flower budding rate. The application of UVA increased the soluble sugar content, whereas higher flavonoid content and antioxidant capacity (FRAP) and lower nitrate content were only observed in Chinese kale exposed to UV6 treatment. Besides, the qPCR assay showed that supplemental UVA-radiation exposure up-regulated the gene expressions of UVR8, transcription factors genes and genes related to the glucosinolate biosynthesis pathway, thereby promoting the accumulation of glucosinolates. Therefore, supplemental UVA-radiation exposure for 12 h/d was more conducive to plant growth, while supplemental UVA-radiation exposure for 6 h/d was better for phytochemical biosynthesis in Chinese kale in an artificial-light plant factory.

The interactive effect of aromatic amino acid composition on the accumulation of phenolic compounds and the expression of biosynthesis-related genes in Ocimum basilicum.

Sweet basil (Ocimum basilicum L.), a well-known medicinal and aromatic herb, rich in essential oils and antioxidants (contributed by phenolics), is widely used in traditional medicine. The biosynthesis of phytochemicals occurs via different biochemical pathways, and the expression of selected genes encoding enzymes involved in the formation of phenolic compounds is regulated in response to environmental factors. The synthesis of the compounds is closely interrelated: usually, the products formed in the first reaction steps are used as substrates for the next reactions. The current study attempted a comprehensive overview of the effect of aromatic amino acid composition (AAAs) in Ocimum basilicum in respect to the expression of genes related to the biosynthesis of phenolic compound and their content. The transcript expression levels of EOMT, PAL, CVOMT, HPPR, C4L, EGS, and FLS increased depending on the AAAs concentration compared to the control plants. The highest mRNA accumulation was obtained in EOMT, FLS, and HPPR in the leaves of sweet basil. The expression of the TAT gene in the leaves significantly reduced in response to all AAAs applications compared to untreated groups and it had the lowest transcript accumulation. Eleven individual phenolic compounds were determined in the basil leaves, and the contents of chicoric acid, methyl chavicol, caffeic acid, and vanillic acid increased depending on administered concentration to control (p < 0.05). Additionally, AAAs lead to an incremental change in the amount of chlorogenic acid at 50 and 100mgkg-1 compared to control plants (p < 0.05). Rutin and rosmarinic acid were detected as the main phenolic compounds in all experimental groups of sweet basil in terms of quantity. However, their amount significantly decreased as compared to control plants based on the increase in AAAs concentrations (p < 0.05). Also, the accumulation of cinnamic acid, eugenol, and quercetin did not significantly change in the leaves of AAAs treated plants compared to control (p < 0.05). When AAAs was applied, total flavonoid content increased in all treatments compared to the control plants, but total phenolic content did not change significantly (p < 0.05). To the best of our knowledge, our work is the first detailed work to evaluate in detail the impact of AAAs on individual phenolic compounds at the phytochemistry and transcriptional levels in the O. basilicum plant. For a detailed understanding of the whole mechanism of phenolic compound regulation, further research is required to fill in some gaps and to provide further clarification.

Full-length transcriptome analysis of pecan (Carya illinoinensis) kernels

Pecan is rich in bioactive components such as fatty acids (FAs) and flavonoids and is an important nut type worldwide. Therefore, the molecular mechanisms of phytochemical biosynthesis in pecan are a focus of research. Recently, a draft genome and several transcriptomes have been published. However, the full-length mRNA transcripts remain unclear, and the regulatory mechanisms behind the quality components biosynthesis and accumulation have not been fully investigated. In this study, single-molecule long-read sequencing technology was used to obtain full-length transcripts of pecan kernels. In total, 37,504 isoforms of 16,702 genes were mapped to the reference genome. The numbers of known isoforms, new isoforms, and novel isoforms were 9013 (24.03%), 26,080 (69.54%), and 2411 (6.51%), respectively. Over 80% of the transcripts (30,751, 81.99%) had functional annotations. A total of 15,465 alternative splicing (AS) events and 65,761 alternative polyadenylation events were detected; wherein, the retained intron was the predominant type (5652, 36.55%) of AS. Furthermore, 1894 long noncoding RNAs and 1643 transcription factors were predicted using bioinformatics methods. Finally, the structural genes associated with FA and flavonoid biosynthesis were characterized. A high frequency of AS accuracy (70.31%) was observed in FA synthesis-associated genes. This study provides a full-length transcriptome data set of pecan kernels, which will significantly enhance the understanding of the regulatory basis of phytochemical biosynthesis during pecan kernel maturation.

UV-C mediated accumulation of pharmacologically significant phytochemicals under light regimes in in vitro culture of Fagonia indica (L.)

Fagonia indica (L.) is an important medicinal plant with multitude of therapeutic potentials. Such application has been attributed to the presence of various pharmacological important phytochemicals. However, the inadequate biosynthesis of such metabolites in intact plants has hampered scalable production. Thus, herein, we have established an in vitro based elicitation strategy to enhance such metabolites in callus culture of F. indica. Cultures were exposed to various doses of UV radiation (UV-C) and grown in different photoperiod regimes and their impact was evaluated on biomass accumulation, biosynthesis of phytochemicals along antioxidant expression. Cultures grown under photoperiod (16L/8D h) after exposure to UV-C (5.4 kJ/m2) accumulated optimal biomass (438.3 g/L FW; 16.4 g/L DW), phenolics contents (TPC: 11.8 μgGAE/mg) and flavonoids contents (TFC: 4.05 μgQE/mg). Similarly, HPLC quantification revealed that total production (6.967 μg/mg DW) of phytochemicals wherein kaempferol (1.377 μg/mg DW), apigenin (1.057 μg/mg DW), myricetin (1.022 μg/mg DW) and isorhamnetin (1.022 μg/mg DW) were recorded highly accumulated compounds in cultures at UV-C (5.4 kJ/m2) dose than other UV-C radiations and light regimes.. The antioxidants activities examined as DPPH (92.8%), FRAP (182.3 µM TEAC) and ABTS (489.1 µM TEAC) were also recorded highly expressed by cultures under photoperiod after treatment with UV-C dose 5.4 kJ/m2. Moreover, same cultures also expressed maximum % inhibition towards phospholipase A2 (sPLA2: 35.8%), lipoxygenase (15-LOX: 43.3%) and cyclooxygenases (COX-1: 55.3% and COX-2: 39.9%) with 1.0-, 1.3-, 1.3- and 2.8-fold increased levels as compared with control, respectively. Hence, findings suggest that light and UV can synergistically improve the metabolism of F. indica and could be used to produce such valuable metabolites on commercial scale.

Nature's Chemists: The Discovery and Engineering of Phytochemical Biosynthesis.

Plants produce a diverse array of natural products, many of which have high pharmaceutical value or therapeutic potential. However, these compounds often occur at low concentrations in uncultivated species. Producing phytochemicals in heterologous systems has the potential to address the bioavailability issues related to obtaining these molecules from their natural source. Plants are suitable heterologous systems for the production of valuable phytochemicals as they are autotrophic, derive energy and carbon from photosynthesis, and have similar cellular context to native producer plants. In this review we highlight the methods that are used to elucidate natural product biosynthetic pathways, including the approaches leading to proposing the sequence of enzymatic steps, selecting enzyme candidates and characterizing gene function. We will also discuss the advantages of using plant chasses as production platforms for high value phytochemicals. In addition, through this report we will assess the emerging metabolic engineering strategies that have been developed to enhance and optimize the production of natural and novel bioactive phytochemicals in heterologous plant systems.

Exogenous application of salicylic acid and gibberellic acid on biomass accumulation, antioxidant and anti-inflammatory secondary metabolites production in multiple shoot culture of Ajuga integrifolia Buch. Ham. ex D.Don

Ajuga integrifolia Buch. Ham. ex D.Don is pharmacologically important plant, commonly known as Kori Booti. Phytochemicals of this plant are used to treat variety of illnesses, ranging from mild tooth ache to malaria and inflammatory diseases. In present study, exogenous role of plant signaling molecules such as salicylic acid (SA) and gibberellic acid (GA) were investigated for enhanced phytochemistry and biological activities in multiple shoot culture of A. integrifolia. Shoots were exposed to various concentrations of SA and GA, and data was collected after 21 days. Significant increase in biomass (dry weight (DW): 17.9 g/L) was observed in response to GA (5.0 mg/L) as compared with SA and control. High performance liquid chromatography was employed to quantify eleven major secondary compounds. Results indicated that salicylic acid promoted phytochemical production in shoot cultures whereas gibberellic acid down regulated their biosynthesis. Salicylic acid (150 μM) was found optimum for maximum accumulation of majority of compounds, while rosmarinic acid and caffeic acid levels were higher in response to SA (75 μM) and SA (300 μM), respectively. In vitro antioxidant and anti-inflammatory activities were significantly increased under SA (150 μM) treatment suggesting a direct correlation in phytochemical biosynthesis and their respective biological activities.

In silico Characterization of microRNAs and Their Target Transcripts from Cranberry (Vaccinium macrocarpon)

MicroRNAs (miRNAs) are highly conserved, non-coding, 20–24 nucleotides long RNA molecules that play important regulatory roles in plants and animals. Due to several limitations involved in the experimental validation of potent miRNAs, in silico prediction of miRNAs and their target(s) from various organisms have been successfully employed. Cranberries are one of the healthiest fruits due to their high nutrient and antioxidant contents. In this study applying genome-wide computational-based approaches and following a set of strict filtering criteria a total of 23 potentially conserved microRNAs belonging to 15 families were identified from cranberry. All the precursors of identified miRNAs formed stable minimum free energy (MFE) stem-loop structure as their orthologues form and possessed high minimum free energy index (MFEI) values. psRNATarget tool detected a total of 92 potential miRNA targets including binding proteins, transcription factors, kinases that are involved in biosyntheses, different metabolic processes, and signal transduction. Among the detected targets, 9 targets (SPLs, proline-rich family proteins, F-Box proteins, HD proteins, Scarecrow proteins, zinc finger proteins, cytochrome P450, sulfate transporters and ABC transporters) were found to have a specific role in phytochemical biosynthesis. To the best of our knowledge, this is the first report of cranberry microRNAs and their targets.

Impact of Copper Oxide Nanoparticles on Enhancement of Bioactive Compounds Using Cell Suspension Cultures of Gymnema sylvestre (Retz.) R. Br

Gymnema sylvestre is a plant that is enriched in bioactive compounds. In particular, gymnemic acids (GA) and phenolic compounds (PC) are pharmaceutically important. There is a commercial demand for naturally occurring bioactive compounds, but their availability is limited due to geographical and seasonal variations. The elicitation approach can enhance the biosynthesis of phytochemicals during in vitro culture of G. sylvestre. Here, to further improve gymnemic acid II (GA II) and phenolic compounds (PC) production by G. sylvestre, cell suspension cultures (CSC), which has attracted attention for the production of essential phytochemicals, was explored using copper oxide nanoparticles (CuO NPs). Callus was obtained on MS medium containing 2,4-dichlorophenoxyacetic acid, kinetin, phytoagar, and sucrose. Agar-free MS medium was used to initiate CSC, which was treated with three concentrations of CuO NPs (1, 3 or 5 mg/L). Treatment for 48 h with 3 mg/L CuO NPs resulted in the greatest yields of GA II, total phenolics, and flavonoids. The cultures also displayed pronounced antioxidant, antidiabetic, anti-inflammatory, antibacterial, antifungal, and anticancer activities. The use of CuO NPs (3 mg/L) significantly increased the production of GA II (nine-fold) and PC compared to unamended CSC. We propose that CSC and use of nanoparticles (NPs) as a new generation of elicitors, offer a suitable prospect for the production of bioactive compounds.

Influence of plant growth regulators on key‐coding genes expression associated with phytochemicals biosynthesis and antioxidant activity in soybean (Glycine max (L.) Merr) sprouts

SummaryThe present study was aimed to investigate the effect of plant growth regulators (PGRs) on phytochemicals biosynthesis and antioxidant activity in soybean sprouts. GmVTC2, GmDHAR and GmGLDH were investigated for the biosynthesis of vitamin C. GmPAL, GmCHS and GmIFS were investigated for the biosynthesis of phenolics, flavonoids and isoflavones. Our results revealed that 6‐benzylaminopurine (6‐BA) treatment significantly affected the expression levels of those major genes, followed by Gibberellin (GA3) and Naphthaleneacetic acid (NAA). Moreover, the highest levels of vitamin C, polyphenolics and antioxidant activity in 6‐BA and GA3 treated groups indicate the synergistic and antagonistic role of growth regulators in the biosynthesis of phytochemicals. Overall, 6‐BA seems the key contributor in genes’ expression involve in the biosynthesis secondary metabolites and their antioxidant activity. Therefore, it could be the most suitable option to enhance nutritional quantity and health benefits of soybean sprouts.

Elicitation of Stevia Glycosides Using Salicylic Acid and Silver Nanoparticles Under Callus Culture

Development of biosynthesis of phytochemicals, especially medicinal products, is highly important due to their broad bioactivity properties. In this study, optimization of callus growth was initially carried out using various combinations of plant growth regulators. Callus with the highest fresh weight was produced on Murashige and Skoog (MS) medium containing 1 (mg/l) naphthalene acetic acid + 0.5 (mg/l) benzyl aminopurine. The effect of different concentrations of salicylic acid (SA) (0.25, 0.5 and 0.75 mg/l) and silver nanoparticles (Ag NPs) (15, 30, 45 and 60 mg/l) on callus growth as well as the possibility of stevia glycosides (SGs) production in callus culture was subsequently evaluated. The SA elicitation, at a concentration of 0.75 (mg/l), resulted in the highest level of callus growth rate (0.1 cm/day), callus diameter (0.79 cm) and relative callus fresh weight (0.085). Likewise, 45 (mg/l) of Ag NPs led to the highest amount of stevioside (32.34 mg/g dry weight callus). The addition of 0.25 (mg/l) of SA to the MS medium led to production of the highest amount of rebaudioside A (3.40 mg/g dry weight callus). The results of this study may enhance the commercial application of important glycosides prevalent in Stevia by highlighting that the nano-elicitors and SA should be utilized at optimized concentrations.

Chitosan elicitation of Isatis tinctoria L. hairy root cultures for enhancing flavonoid productivity and gene expression and related antioxidant activity

Elicitation for phytochemical enhancement via cost-effective elicitors can overcome the limitation of commercial application faced by plant cell and organ culture technology. Chitosan is a natural, low-cost, and nontoxic elicitor that can trigger plant defense responses with the concomitant enhancement in phytochemical biosynthesis. In this work, the elicitation of Isatis tinctoria L. hairy root cultures by chitosan was conducted to enhance the production of pharmacologically active flavonoids. In comparison with control (2.31 ± 0.29 mg/g DW), a 7.08-fold enhancement of total flavonoids (16.35 ± 0.88 mg/g DW) was achieved in 24 day-old I. tinctoria hairy root cultures elicited by 150 mg/L chitosan for 36 h. Interestingly, the multiple hydroxyl-substituted flavonoids (rutin, quercetin, isorhamnetin, and isoliquiritigenin) were noticed to increase significantly in chitosan-elicited I. tinctoria hairy root cultures. Moreover, the transcription of associated genes involved in flavonoid biosynthesis pathway was significantly up-regulated underlying chitosan elicitation, among which chalcone synthase and flavonoid 3′-hydroxylase might play an important role in flavonoid enhancement. Additionally, extracts from chitosan-elicited I. tinctoria hairy root cultures exhibited higher antioxidant activities with lower IC50 values as compared with control. Overall, a cost-effective strategy via the simple chitosan elicitation is provided here to enhance the production of high-added value flavonoids in I. tinctoria hairy root cultures, which paves the way toward the successful commercialization of this in vitro culture system in the future.

Effects of simultaneous UV-C radiation and ultrasonic energy postharvest treatment on bioactive compounds and antioxidant activity of tomatoes during storage

The effects of a novel technology utilizing a simultaneous combination of Ultraviolet-C radiation and ultrasound energy postharvest treatment on tomato bioactive compounds during 28 days’ storage period was investigated by varying Ultraviolet-C radiation intensities of 639.37 or 897.16 µW/cm2 at a constant ultrasound intensity of 13.87 W/L from a 40 kHz–1 kW transducer. A minimal treatment time of 240 s at Ultraviolet-C dosage of 2.15 kJ/m2 was observed to provoke a considerable increase in bioactive compounds content, proportionated to treatment time. Although treatment led to temperature increase in the system reaching 39.33 °C due to heat generation by ultrasonic cavitation, the extractability and biosynthesis of phytochemicals were enhanced resulting in 90%, 30%, 60%, 20%, and 36% increases in lycopene, total phenols, vitamin C, hydrophilic and lipophilic antioxidant activities respectively. Results present the potential use of the combined non-thermal technologies as post-harvest treatment to improve bioactive compounds and antioxidant activity during storage.

De Novo Transcriptome Profiling of Buasbuas (Premna pubescens. Blume)

Buasbuas is the one of the medicinal plants in Indonesia that contains bioactive compounds potential as antimicrobial, antioxidant, antidiabetes, antiinflammation and anticancer. Exploring the pathway and gene related of buasbuas bioactive compounds production has led to the renaissance of understanding buasbuas molecular mechanism database. The aim of this study was to developed data-mining framework of buasbuas to study plant specialized metabolism for phytochemical biosynthesis. Material and Methods: This project was started by collecting shoots and leaves of Buasbuas. Focus of the project was exploring the molecular mechanisms on biosynthesis phytochemical of Buasbuas. Illumina Mi-Seq Next Generation Sequencing was utilized to understand the molecular mechanisms of biosynthesis. Transcriptomes then trimmed and assembled with CLCBio genomic software. Assembled contigs then annotated towards Arabidopsis thaliana using CLC Bio genomic software. Digital Gene Expression was performed to analyze the transcriptional changes in control culture and treatment. Results: There were 5.342 unigenes that expressed only in treatment shoot cultures. Annotation with Gene Ontology showed that 57.9% (3.446) unigenes play role in Biological Process, 56.7% (3.375) unigenes play role in Cellular Components and 63.4% (3.772) unigenes play role in Molecular Functions. Annotation with Kyoto Encyclopedia of Genes and Genomes shows 853 unigenes essentially have role in 24 biological pathways. Highest process with highest unigenes involvement is biosynthesis of plant hormones and biosynthesis of alkaloids. Conclusion: This study showed that phytochemical biosynthesis in buasbuas induces level expression of several genes involved in the jasmonic acid, cytokinin, gibberellin, salicylic acid and ethylene biosynthesis pathway.
